体外研究 去甲肾上腺素(NE)酒石酸氢盐一水合物通常被认为是β1亚型选择性肾上腺素能激动剂。去甲肾上腺素(NE)在β2-肾上腺素能受体中也具有更高浓度的直接活性[1]。从新生儿野生型C57BL/6J小鼠中分离来自腹股沟脂肪垫(iWA)或肩胛间脂肪垫(BA)的脂肪细胞并进行培养。为了检查激活AT2对β-肾上腺素能信号传导的影响,首先评估cAMP产生以响应去甲肾上腺素(NE,10μM),有或没有CGP(10nM)共处理。去甲肾上腺素(NE)如iWA中预期的那样增加cAMP,并且CGP不会改变这种效应。还已知去甲肾上腺素(NE)诱导脂解,并且需要释放的脂肪酸来功能性激活UCP1蛋白并刺激产热。在去甲肾上腺素(NE)治疗后,Ser133处的CREB磷酸化增加,并且在小鼠iWA中通过CGP共处理显着减弱[2]。
细胞实验 来自38岁非糖尿病女性供体的皮下前脂肪细胞用TERT和HPV E6 / E7永生化。对于目前的研究,通过环克隆分离具有一致分化能力的稳定二倍体克隆(称为克隆B)。细胞在前脂肪细胞PGM2培养基中生长。一旦细胞汇合,通过在由地塞米松,IBMX,吲哚美辛和另外的胰岛素组成的分化培养基中孵育诱导分化。细胞分化10天。在处理之前,将培养基替换为PGM2培养基一天,然后切换至无血清培养基过夜进行处理。用载体,去甲肾上腺素(NE,10μM),CGP(10nM)或去甲肾上腺素(NE)和CGP [2]处理脂肪细胞6小时。
