| 体外研究 | (+)-盐酸二氢西汀(DHX)是一种高效,生物可利用的D1多巴胺受体激动剂。 (+)-筛选二氢西汀对40个其他结合位点的活性,并对除D2多巴胺受体(IC50 = 130nM)和α2肾上腺素受体(IC50 = 230nM)以外的所有部分无活性(IC50大于10μM)。二氢哌啶立体选择性地和有效地竞争用[3H] SCH23390标记的大鼠纹状体膜中的D1结合位点,IC50为约10nM,而原型D1激动剂SKF38393为约30nM [2]。 |
| 体内研究 | 为了检查纹状体神经元响应D1受体激活的功能状态,AC5 +/+和AC5 -/-小鼠注射D1激动剂(+)-二氢西汀(30 mg/kg,ip)并获得背外侧纹状体。分别在45分钟后进行RT-PCR分析和NAc。这些实验表明,与AC5 +/+小鼠相比,AC5 -/-小鼠中(+)-二氢西汀对NAc中立即早期基因c-fos,egr-1和junB的诱导显着增强,而在AC5 -/-小鼠中,背外侧纹状体的诱导被抑制[3]。 |
