体外研究 类固醇硫酸酯酶-IN-2(化合物1,约0-20μM,72小时)抑制不同的癌细胞,IC50范围为1.26μM至17.86μM[1]。类固醇硫酸酯酶-IN-2(1μM,5小时)释放多达四个一氧化氮分子[1]。类固醇硫酸酯酶-IN-2(8μM,8-72小时)通过MAPK途径诱导白血病细胞CCRF-CEM凋亡,将细胞阻滞在G2/M期,并增加Bax/Bcl-2的比率[1]。凋亡分析[1]细胞系:CEM细胞浓度:8μM孵育时间:8、24、48或72小时结果:诱导凋亡呈时间依赖性和剂量依赖性。细胞周期分析[1]细胞系:CEM细胞浓度:8μM孵育时间:8、24、48或72小时结果:细胞阻滞于G2/M期。Western Blot分析[1]细胞系:CEM细胞浓度:0-8μM孵育时间:72小时结果:增加JNK和p38的水平,以及磷酸化JNK的水平和p38。以时间和剂量依赖性方式降低Bcl-2水平,并增加促凋亡Bax水平。
