由甘氨酸缩合而得。将甘氨酸与乙二胺按1：5混合后搅拌加热，174-176℃反应50min，然后置于冰室中过认，过滤出结晶，用甲醇洗涤，干燥得甘氨酰酐。也可以由甘氨酸在脱水的甘油中缩合而制备。
