以丙烯腈为原料，在碱性条件下与氰氢酸反应制得。收率95%以上。也可将氰化钠水溶液加热，滴加1,2-二溴乙烷的乙醇溶液，回流反应35h，可生成丁二腈。

精制方法：将丁二腈进行升华精制。或用丙酮重结晶后进行减压蒸馏，可得高纯度产品。
