[ 描述 ]:

Win 18446 是具有口服活性的睾丸特异性的酶 ALDH1a2 的抑制剂,其 IC50 值为 0.3 μM。Win 18446 可逆的抑制多个物种的精子形成,抑制睾丸内视黄醇 (HY-B1342) 合成视黄酸 (HY-14649)。

[体外研究]

WIN 18446(1μM,24 h)能够在含有生殖细胞和体细胞的整个睾丸培养物中抑制Stra8的表达[2]。RT-PCR[2]细胞系:取2-dpp小鼠睾丸。浓度:1μM,孵育时间:24h。结果:睾丸呈VAD样表型。单独或联合使用ROL可显著降低Stra8的表达。

[体内研究]

Win 18446(200mg/kg,口服4、8和16周)显著降低睾丸内维甲酸浓度,严重损害精子发生,并导致不育[1]。动物模型:6月龄雄性新西兰大白兔[1]。剂量:200mg/kg。给药:口服4、8、16周。结果:随着时间的推移,睾丸重量明显减轻。显著抑制精子发生。显著降低睾丸内维甲酸浓度和Stra8表达,严重抑制精子发生和生育。
