[ 靶点 ]

Ki: 18.9 nM (Human A3 receptor), 113 nM (Rat A3 receptor), 15.6 µM (A1 receptor) and 2.05 µM (A2A receptor)[1]

[体外研究]

MRS 1523(0.1-1μM)处理能显著拮抗细胞数,分别达到对照值的40.7%和57.4%,在加入虫草素(60μM)前30min。单用MRS1523(1μM)对肿瘤细胞生长有影响[3]。当人内皮祖细胞(hEPC)与MRS 1523(1nm)共孵育时,观察到部分阻断腺苷-5'-N-乙基羧酰胺(NECA)诱导的迁移。此外,在3天的hEPC中,MRS 1523100nm抑制NECA诱导的迁移70%。MRS 1523以剂量-反应方式阻断NECA诱导的迁移,计算Ki为147nm[4]。细胞活力测定[3]细胞株：B16-BL6细胞浓度：0.1μM,1μM培养时间：24小时、48小时、72小时结果：对抗虫草素诱导的生长抑制。

[体内研究]

研究了A3腺苷受体(A3AR)在3～4周龄大鼠中小型辣椒素敏感背根神经节(DRG)神经元兴奋性中的表达及功能作用。内源性激动剂腺苷减少N型钙电流,在A3AR拮抗剂MRS 1523存在下,其作用被抑制56%。电流钳记录显示,大鼠DRG神经元的放电也明显减少了A3AR激活,MRS 1523敏感,PD173212不敏感。
