O-Phospho-L-serine 是 L-serine 合成过程中的直接前体物质,是第三组 mGluR (mGluR4, mGluR6, mGluR7, and mGluR8) 的激动剂,O-Phospho-L-serine 同时为 mGluR1 和 mGluR2 的拮抗剂。

[体外研究]

O-磷酸-L-丝氨酸(1-SOP)与mGluR1弱结合,并拮抗1-谷氨酸的作用。 l-SOP激活III组受体(mGluR4,mGluR6,mGluR7和mGluR8),但mGluR7对l-SOP的亲和力低于其他III组受体,并且对两种配体的效力也较低[1]。 O-磷酸-L-丝氨酸(1-SOP)在mGluR4转染的细胞中产生增强的细胞内钙响应。 l-SOP抑制l-谷氨酸介导的mGluR1反应,Ki为1 mM; l-SOP显示出对mGluR2活化的显着更强的抑制,Ki为1μM,比mGluR1更有效三个数量级。 l-SOP诱导用mGluR4或mGluR6转染的HEK/TRPC4细胞中的膜电位变化。 l-SOP诱导由Gαi/ o蛋白介导的TRPC4β活化[2]。 O-磷酸-L-丝氨酸(L-SOP)抑制Müller胶质细胞增殖,而不影响光诱导的光感受器细胞死亡。 L-SOP在光损伤视网膜中激活ascl1a和stat3表达之后或与之并行地破坏Müller胶质细胞增殖。 L-SOP抑制光损伤视网膜中的视锥细胞再生[3]。
