以L-半胱氨酸与醋酐乙酰化即得
中和 将无结晶水的L-半胱氨酸盐酸盐溶于0.8倍的蒸馏水中,经水浴微热溶解后倒人预先铺有活性炭的布氏漏斗中抽滤,取滤液并加入乙醇 (95%),再加入吡啶搅拌后,放人冰水浴中冷却,再置人冰箱中过夜,析出结晶,过滤取结晶,用CHCl3洗涤,压干。迅速放人真空干燥器中干燥,得L-半胱氨酸。
乙酰化 将L-半胱氨酸投入反应罐中,加入冰醋酸、三氯甲烷,在剧烈搅拌下加入醋酐。反应罐夹层加热至35℃左右后,停止加热,罐内温度逐渐上升至82-85℃,为控制温度过高,在70℃左右时开始冷却,维持50℃左右搅拌半小时后,过滤取滤液,并减压浓缩,当比较黏稠时,再加适量水浓缩。如此反复几次,当浓缩液为半胱氨酸1.8倍左右时,冷却并加入少许L-半胱氨酸晶体接种,放入冰箱中过夜、结晶,过滤取结晶压干,用乙醚洗涤,得乙酰半胱氨酸粗品。
精制 粗品放入0.5-0.6倍的蒸馏水中溶解,加5%活性炭脱色,过滤取滤液,放人冰箱中过夜,析出结晶,过滤取结晶,压干,用乙醚洗涤,真空干燥,得乙酰半胱氨酸精品。
