[体外研究]

向含有0.25μM3,3'-二己氧基羰花青菁碘化物(DiOCg(3))的比色皿中添加细胞导致荧光增加,平衡快速并且完成4分钟。当将f-met-leu-phe(10-7M)加入到用3,3'-二己氧基羰花青菁碘化物预平衡的嗜中性粒细胞中时,存在5秒的短暂滞后期。滞后期之后是荧光的快速丧失。使用荧光显微镜检查负载3,3'-二己氧基羰花青素的嗜中性粒细胞表明,在静息中性粒细胞中,最亮的荧光与细长的细胞器有关,这与细胞分布相对应[1]。

[细胞实验]

将中性粒细胞（2.5×10 6 / mL）在0.25μM3,3'-二己氧基羰花青菁碘化物（DiOCg（3））中在37℃下在1cm路径长度的比色皿中预平衡5分钟。最终体积为2 mL。荧光测量在荧光分光光度计中进行（发射波长：510nm，激发波长：470nm）。通过注射器通过分光荧光计上的注射口注射刺激物，其体积不超过5％（v / v）[1]。
