[体外研究]

GW4869(10μM)部分抑制TNF诱导的鞘磷脂(SM)水解,并且20μM化合物完全免于SM的损失。添加10-20μMGW4869完全抑制神经酰胺的初始积累,而这种效应在稍后的时间点(24小时)部分丧失。 GW4869的作用发生在谷胱甘肽下降的下游。 GW4869能够以剂量依赖性方式显着防止细胞死亡。这些保护作用伴随着线粒体和胱天蛋白酶9活化的细胞色素c释放的显着抑制,因此在线粒体功能障碍的上游定位N-SMase活化[1]。

[体内研究]

用GW4869预处理显着损害RAW264.7巨噬细胞中外来体和促炎细胞因子(TNF-α,IL-1β,IL-6)的释放。在LPS处理或CLP手术后12小时,用GW4869预处理的WT小鼠在血清中显示出比对照PBS注射小鼠更低量的外泌体和促炎细胞因子。因此,GW4869治疗可减少脓毒症引起的心脏炎症,减轻心肌抑制并延长生存期[2]。

[激酶实验]

B. cereu sphosphatidylcholine-PLC在存在或不存在10μMGW4869的情况下在含有100mM Tris，pH7.2,25％甘油，20mM对硝基苯基/磷酰胆碱的反应混合物中温育，并且通过分光光度法定量生产对硝基苯酚在410纳米。将来自牛肾的蛋白磷酸酶2A在GW4869存在或不存在下在含有50mM Tris，pH7.4,1mM二硫苏糖醇，100μMMnCl2和20％甘油的缓冲液中温育，并测量磷酸酶活性[1]。

[细胞实验]

用GW4869处理细胞30分钟，然后以10μL/孔加入TNF。在指定的时间点，向每个孔中加入25μLMTT储备溶液，并在37℃，5％CO 2中孵育3小时。细胞活力是使用MTT测定[1]。

[动物实验]

小鼠：将小鼠随机分成四组：PBS，GW4869，PBS + LPS和GW4869 + LPS（每组n = 5）。 GW4869以2.5μg/ g的一个剂量腹膜内（ip）注射。在ip注射LPS（25μg/ g）之前1小时腹膜内预注射PBS + LPS组中的小鼠。将GW4869 + LPS组中的小鼠腹膜内注射GW4869（2.5μg/ g）1小时，然后腹膜内注射LPS（25μg/ g，100μl）。小鼠接受PBS注射至与对照相当的体积（100μl）。每6小时监测PBS + LPS和GW4869 + LPS组的存活率，持续36小时[2]。
