[体外研究]

钙黄绿素AM仅是弱荧光(激发/发射最大值322/435nm)。在通过细胞内酯酶裂解AM酯时,该示踪剂变得相对极性并被细胞保留数小时。此外,其荧光强度增加并且其荧光光谱向更长波长移动,激发/发射最大值为360/449nm。已知钙黄绿素的荧光在铁(III)存在下猝灭;如果更强的螯合剂从荧光团中除去这种离子,荧光团的荧光就会重新获得[1]。描述了使用与Fe 2离子络合的钙黄绿素(CB)作为化学传感器的新型多巴胺荧光测定法。由钙黄绿素-Fe2复合物的钙黄绿素产生的荧光被Fe2离子猝灭。当将多巴胺加入到钙黄绿素-Fe2复合物的溶液中时,由于钙黄绿素和多巴胺之间的配体交换反应,形成多巴胺-Fe2复合物,这使得钙黄绿素的荧光得以恢复。发现波长为440nm(在340nm的激发波长下)的荧光强度与加入到钙黄绿素-Fe2复合物溶液中的多巴胺的浓度成比例,这允许定量测定多巴胺[2]。

[细胞实验]

建议的最终工作浓度通常在1到10μM之间，以尽量减少潜在的伪影。通常，15分钟至1小时足以用于细胞摄取和染料的加工。在0.1M氢氧化钾中制备5mM 4-甲基伞形酮-8-甲基亚氨基二乙酸的储备浓度，通常称为钙黄绿素，并用0.1N盐酸中和pH。将制备的钙黄绿素原液在DPBS中稀释至200μM（工作原液）并储存以供进一步使用。通过在DPBS中稀释，从工作原料制备浓度为10μM的钙黄绿素蓝。由此，将200μL添加到96孔板中，并使用微板读数器[1]确定激发/发射（Ex./Em。）最大值。
