[体外研究]

溶液中的溴代溴化物(中等极性的水性或有机溶剂)与小硫醇[例如,三肽硫醇谷胱甘肽(GSH)]和反应性蛋白质硫醇基团(例如,血红蛋白)反应。溴代二甲双胍与硫醇的反应是二级的并且取决于pH,活性亲核试剂是硫醇盐阴离子。溴代二甲双胍与硫醇盐的反应将非荧光剂转化为水溶性荧光产物。中性试剂mBBr和bBBr适度溶于中极性有机溶剂(乙腈,二氯甲烷),微溶于水。季盐qBBr和阴离子溴代双环硼烷SBBr可溶于水,但在有机溶剂中溶解性较差。 Bromobimanes是黄色的[1]。溴代二甲双胍对硫醇的高选择性,快速反应性,硫醇衍生物的稳定性和荧光产率,反相高效液相色谱法分离衍生物的容易性,以及细胞穿透和非渗透形式的可用性,都可以使用溴代溴醚一种分析低分子量生物硫醇的极其有效的方法[2]。

[细胞实验]

原料mBBr溶液按重量在100-180mM乙腈中制备。 SBBr的储备溶液在二甲基亚砜（DMSO）中以50mM的重量或在20mM的Tris-甲磺酸盐中以2mM的重量制备。将硫醇加至终浓度为1mM至2mM mBBr或SBBr，20mM Tris-甲磺酸盐，pH8.0的溶液中。将混合物在室温下在黑暗中温育15分钟，然后从5M储备溶液中加入甲磺酸至25mM [2]。
