[体外研究]

甲基萘醌-4(MK-4,0,1,5,10μM)增加Caco-2细胞中的ALP活性。甲基萘醌-4(1μM)显着增加hSI表达的强度[1]。

[体内研究]

HF-K2组中的甲基萘醌-4(K2,0.2g/kg饮食)在C57BL/6J小鼠中产生附睾脂肪,并且还增加小鼠的骨密度[2]。

[细胞实验]

将Caco-2细胞以2-5×10 4个细胞/ cm 2的密度接种到35-mm培养皿上。将细胞温育2至3天直至60%-70%汇合,并加入所需浓度的甲基萘醌-4(0,1.0,5.0和10.0μM)。载体的最终浓度为培养基的0.1%,培养基每周更换两次。在添加Menaquinone-4 [1]后第0,3,7和11天测定细胞。

[动物实验]

42只雄性,4周龄的C57BL / 6J小鼠随意提供饲料和饮料。对于环境适应,动物有1周的循环,然后在随机分成6组(每组7只动物;随机区组设计)后提供实验饮食。 AIN-93G饮食包括正常饮食(N),正常饮食+维生素K1(N-K1),正常饮食+维生素甲萘醌-4(N-K2),45%高脂饮食(HF),45%高脂肪饮食+维生素K1(HF-K1),45%高脂饮食+维生素甲萘醌-4(HF-K2)。维生素K1和维生素甲基萘醌-4含量为200mg / 1,000g,饮食以颗粒形式提供。每周测量一次体重,并且通过将实验期间的食物摄入量除以第1天至最后一天的增加的体重来计算食物效率比(FER)。对于脂肪量测量,从死亡动物受试者中提取附睾脂肪,肾周脂肪和腹膜后脂肪,用0.9%NaCl洗涤,用滤纸干燥,然后称重[2]。
