• 体外研究: 干细胞再生素1(SR1)通过拮抗芳烃受体(AhR)起作用。干细胞再生素1在5至7天后增加CD34 +细胞的数量,EC50为~120nM。 StemRegenin 1抑制光亲和配体(PAL)结合(IC50 = 40 nM)这些结果支持StemRegenin 1诱导的CD34 +细胞扩增是通过直接结合和抑制AhR介导的结论[1]。芳烃受体拮抗剂StemRegenin 1(SR1)强有力地促进人CD34 +细胞的离体扩增。 StemRegenin 1治疗加速CD34 +细胞的增殖并降低VentX的表达水平[2]。
• 细胞实验: 将一定量的250,000个CB衍生的CD34 +细胞与对照条件（DMSO，0.01％）或StemRegenin 1（0.75μM）一起培养3周。此时，对照培养物扩增了1080倍，StemRegenin 1处理的细胞相对于起始细胞数扩增了2024倍。移植30至30,000个未培养的CD34 + CB衍生细胞或相当于30至10,000个起始细胞的最终培养物的一部分。通过眼眶后路径将细胞静脉注射到亚致死照射的（300拉德，200拉德）6至10周龄的NSG小鼠中。在照射后24小时内进行植入。通过使用抗人CD45和抗小鼠CD45抗体通过眶后窦或骨髓获得的血液的流式细胞术分析来监测植入。在移植后13-16周处死小鼠;收集骨髓（来自股骨和胫骨），脾脏和胸腺用于分析。对于二次植入，将来自每只受体小鼠的50％的骨髓移植到一个次级亚致死照射的NSG小鼠中。移植后15周，从次级小鼠收集骨髓并通过流式细胞术分析[1]。
