• 体外研究: MD-224(1-30 nM; 2小时)有效地诱导MDM2蛋白的耗竭和p53蛋白在RS4; 11细胞中以剂量依赖性方式同时积累[1]。MD-224(30nM; 6小时)在诱导这些p53靶基因的转录上调方面比MI-1061更有效,但对RS4; 11细胞中的TP53本身没有影响[1]。MD-224(0.001-1μM; 24小时)在24小时治疗后以≤10nM的剂量依赖性方式诱导强壮的细胞凋亡[1]。Western Blot分析[1]细胞系：RS4；11细胞浓度：1nM；3nM；10nM；30nM孵育时间：2小时结果：MDM2蛋白降低,p53蛋白积累。RT-PCR[1]细胞系：RS4;11细胞浓度：30nM孵育时间：6小时结果：p53靶基因表达上调。细胞凋亡分析[1]细胞系：RS4;11细胞浓度：0.001μM,0.003μM,0.01μM,0.03μM,0.1μM,0.3μM,1μM培养时间：24小时结果：诱导RS4;11细胞强烈凋亡。
