[体外研究]

Tolfenamic Acid是一种非甾体类抗炎药,选择性抑制COX-2,在LPS处理的(COX-2)犬DH82单核细胞/巨噬细胞中IC50为13.49μM(3.53μg/ mL),但对COX-1没有影响。 [1]。托芬那酸(100μM)抑制BE3,OE33和SKGT5的> 70％的细胞活力。 Tolfenamic Acid还可作为有效的Sp蛋白抑制剂,降低Sp1和Sp4,抑制食管癌细胞BE3和SKGT5中c-Met的表达[2]。 Tolfenamic Acid(50μM)显着影响L3.6pl细胞中的基因表达,并下调CENPF,KIF20A,LMNB1,MYB,SKP2,CCNE2和DDIT3 [3]。

[体内研究]

Tolfenamic Acid(50mg/kg 3次/周,po)抑制N-亚硝基甲基苄胺(NMBA)诱导的食管肿瘤模型中的肿瘤形成和肿瘤发生率。在用NMBA治疗的大鼠中,Tolfenamic Acid也会导致肿瘤多样性和肿瘤体积减少[2]。

[细胞实验]

所有细胞在补充有5％血清的培养基（RPMI1640）中生长，并在37℃，95％空气和5％CO 2的潮湿气氛中培养。接种后24小时，用载体（0.1％DMSO）或各种浓度的托芬那酸（25/50 /100μM）处理细胞。在处理后24,48和72小时进行细胞活力测定。用50μM托芬那酸处理细胞，并在处理后48小时收获细胞沉淀。这些颗粒用于制备用于蛋白质印迹分析的细胞裂解物[2]。

[动物实验]

小鼠[2]食管SCC的Fischer 344（F-344）大鼠模型最初每笼饲养两只，但由于体积增大最终分成每笼一只，并保持在标准的人道条件下（20±2°） C，50±10％相对湿度，12小时光照/黑暗循环）。随意提供食物和水。每次给药时记录体重。到达动物设施后两周，将大鼠随机分成4组：C：NMBA（1-5周）; NTA :( NMBA 1-5周，然后Tolfenamic Acid 6-25周）; NC：阴性对照（载体）;和TA：Tolfenamic Acid阴性对照。对照（C）和NTA组用NMBA（0.5mg / kg）在0.2mL载体中皮下注射，每周三次，持续5周，而阴性对照组仅注射载体。从第6周到第25周，NTA和托芬那酸组也通过口服强饲法接受50mg / kg托芬那酸。在第25周后，处死动物，纵向切开食道，并对表面肿瘤进行定位和计数。记录包括息肉在内的病变的数量和大小并捕获图像。使用长椭球体的公式计算肿瘤体积：长×宽×高×p / 6 [2]。
