[体外研究]

TAS-301(1-10μM)浓度依赖性地抑制由PDGF诱导的PKC活化和Ca2 +内流,对10μM时PKC活化的抑制率为62.7％,并且降低PMA诱导的AP-1,抑制率为38％和67.6％分别为3和10μM[1]。 TAS-301(0.3-3μM)剂量依赖性地减少由生长因子(PDGF-BB,IGF-1,HB-EGF)诱导的细胞迁移。 TAS-301(1-10μM)也可降低bFGF诱导的BrdU掺入,特别是在3和10μM时[2]。

[体内研究]

在球囊损伤后14天,TAS-301(3-100mg/kg,口服)剂量依赖性地降低新内膜增厚和I/M比并降低大鼠内膜细胞水平[2]。

[细胞实验]

通过静止细胞掺入BrdU来确定细胞增殖。将SMC以1×10 4个细胞/孔接种在含有10％FBS的DMEM中的96孔板中。接种后两天，在含有5μg/ mL胰岛素，5μg/ mL转铁蛋白和5ng / mL硒钠（ITS）的无血清DMEM中将其生长停止3天。然后，除去DMEM / ITS，在用生长因子（即，bFGF 0.1ng / mL）处理前2小时，将含有或不含TAS-301或曲尼司特的0.1％BSA的无血清DMEM加入静止细胞中。在刺激后24小时，将BrdU（10μM）加入细胞中; 24小时后，细胞固定。 ELISA用于检测和定量掺入的BrdU（n = 6）。在整个实验期间存在药物[2]。

[动物实验]

大鼠[2]在球囊损伤后第14天，用乙醚麻醉大鼠以避免对动物施加任何压力，然后用盐水经心脏灌注，接着用10％缓冲的福尔马林灌注。接下来，移除左颈动脉（从主动脉弓到分叉的长度），后固定并包埋在石蜡中。然后，切割3μm厚的动脉切片（每个动脉6个切片）并用苏木精和曙红染色。通过使用数字分析仪测量照片上的内膜和介质的横截面积。确定每个动脉中内膜面积与中间面积的比率的平均值。实验组如下：载体（n = 9），TAS-301（3,10,30和100mg / kg，n = 9）和曲尼司特（100和300mg / kg，n = 9）。由于口服给药错误导致死亡，2只大鼠（一只在TAS-301 100 mg / kg组，一只在曲尼司特100 mg / kg组）的数据被省略[2]。
