[体外研究]

Procarbazine Hydrochloride是一种抗癌剂。 Procarbazine对缺乏细胞色素P-450或单胺氧化酶活性的LI 210细胞没有细胞毒性[1]。

[体内研究]

盐酸丙卡巴肼(50mg/kg,ip)在造血细胞中引起微核,但不增加小鼠骨髓中的lacZ突变频率(MF),类似于肝,睾丸,脾,肾和肺中的那些。盐酸丙卡巴肼(50 mg/kg,ip)对肺癌,骨髓和脾脏有致癌作用[2]。 Procarbazine(450 mg/kg)显着降低睾丸和附睾重量,并显着减少单倍体细胞和仓鼠中的生精停滞[3]。

[细胞实验]

在丙卡巴肼或代谢物处理后,将细胞在25-cm 2培养瓶(10mL)中稀释至50.000 / mL。每24小时,取出0.5-mL等分试样,在Hematall等渗稀释剂中稀释20倍,并用Coulter Model F电子细胞计数器测定细胞数。大于10,000 / 0.5 mL的计数校正重合。当细胞密度超过1×106 / mL时,细胞在新鲜培养基中稀释。保持培养物直至聚集细胞数接近100×106 / mL并且倍增时间恢复至12小时。使用方程式A确定细胞存活,其中TD(感兴趣的细胞的倍增时间)是12小时[1]。

[动物实验]

小鼠[2]在适应2周后使用雄性MutaTM小鼠动物(7-8周龄)。在第一个实验中,18只小鼠腹膜内(ip)注射50mg / kg盐酸丙卡巴肼10mL盐水/ kg,8只小鼠注射10mL盐水/ kg作为媒介物对照。处理后7,14和28天处死6只处理过的小鼠,处理后7天和28天处死4只对照小鼠。杀死是由颈椎脱位[2]。
