[体外研究]

离子霉素是一种钙离子载体,是由链霉菌产生的抗生素[1]。向LCLC 103H细胞中添加2μM离子霉素导致细胞内Ca2+浓度从50nM瞬时增加至180nM。离子霉素处理的培养物中的DNA和蛋白质分析显示DNA片段化和PARP裂解为典型的半胱天冬酶介导的细胞凋亡的85-kDa片段。在同时阳性荧光素标记和碘化丙啶摄取的支持下,约有1-5％的离子霉素处理细胞可检测到坏死。随后在整个细胞中激活半胱天冬酶,然后监测离子霉素处理后对Ac-DEVD-amc活性的增加[2]。
