以18-甲基雌甾-4-烯-3,17-二酮为原料，经发酵在15位引入羟基。溶于吡啶，加入甲磺酰氯，反应后再加入二甲基甲酰胺和无水乙酸钠。再在四氢呋喃和己烷的混合液中，正丁基锂存在下，通入乙炔，进行乙炔化，得孕二烯酮。

或者18-甲基雌甾-4-烯-3,17-二酮为原料，经发酵在15位引入羟基。先在吡啶中，用
乙酸酐进行酰化，再进行醚化。然后在四氢呋喃中，正丁基锂存在下，通入乙炔进行乙炔化。最后在盐酸作用下，得到孕二烯酮。
