[体外研究]

Cremophor EL是一种广泛使用的赋形剂。在前10小时,5mg/mL的Cremophor EL增加阻抗,然后降低阻抗,并且在治疗20小时后,检测到上皮细胞死亡。 Cremophor EL比RH40降低细胞活力并增加细胞毒性。在24小时处理时,在10至50mg/mL浓度的Cremophor EL下,上皮活力和单层完整性均严重受损。 Cremophor EL也可以增强细胞分离[1]。

[细胞实验]

通过双荧光染色测定细胞活力：双 - 苯甲酰亚胺标记所有细胞核，乙锭 - 同型二聚体-1仅染色死细胞。对于形态学检查，使用具有紧密屏障特性的原代脑内皮细胞。在玻璃盖玻片上生长的细胞用Cremophor EL和RH40处理1和24小时。 Cremophor EL和RH40在0.1至50mg / mL浓度下进行测试。 Triton X-100去污剂（10 mg / mL）用于毒性试验，作为引起细胞死亡的参考化合物[1]。

聚氧乙烯蓖麻油EL制备

将1 mol蓖麻油加入反应釜中，再加入相当蓖麻油质量的0.3%的固体氢氧化钾。搅拌，升温，至KOH溶解。然后用N2置换釜中空气。并通入环氧乙烷，反应温度维持在160～180 ℃，压力0.15～0.2 MPa。通毕后继续搅拌至压力不再下降。冷却。将物料移入漂白釜，用洋醋酸调pH至微酸性，加0.1%的双氧水脱色，70 ℃下保温0.5 h。最后减压脱水5 h。冷却出料。反应式如下。
