透明质酸钠制备
是透明质酸羧基与金属钠离子所成的盐。透明质酸普遍存在于动物和人体内,如皮肤、肌肉、软骨、脑、鸡冠、动脉和静脉壁等。常用提取原料有雄鸡冠、脐带、牛眼等。以鸡冠为原料的制取方法如下。
1.提取:新鲜鸡冠用丙酮洗、脱水粉碎,加蒸馏水浸泡提取24h,重复3次,合并滤液。
2.除蛋白、沉淀:提取液与等体积氯仿混合搅拌3h,分出水相,加2倍量乙醇,收集沉淀、丙酮脱水、真空干燥,得粗品透明酸。
3.再经酶解、络合、解离、沉淀得精品透明质酸。然后和金属钠离子成盐。
