[体外研究]

α-雌二醇(17α-雌二醇)是一种5α-还原酶抑制剂,可抑制5α-还原酶催化的睾酮代谢[1]。 α-雌二醇(17α-雌二醇,10μM)以ERα依赖性方式减弱C57BL/6J雄性和雌性小鼠胚胎成纤维细胞(MEF)细胞,原代前脂肪细胞和分化的3T3-L1脂肪细胞中LPS诱导的炎症标志物。 ,这种作用是通过降低NFκB-p65和增加ERα蛋白表达[2]。

[体内研究]

[动物实验]

根据治疗方式将新生小鼠随机分为6组：室内空气注射媒介物（对照组，第1组），车辆注射高氧（对照组，第2组），高氧用0.01μgα-雌二醇注射（第3组）高氧，0.1μgα-雌二醇注射（第4组），高氧与1.0μgα-雌二醇注射（第5组），高氧与1.0μgα-雌二醇和10.0μgICI182780注射（雌激素受体α和β的拮抗剂）（第6组）。幼仔每天皮下注射载体中的α-雌二醇[溶于乙醇并在0.05mL /小鼠的磷酸盐缓冲盐水（PBS）中稀释]或从出生后天（PND）7-16单独的载体。在PND 7，高氧和α-雌二醇治疗组的幼鼠暴露于高氧（75±2％O2）5天（PND 7-12），然后恢复到常氧（室内空气）5天，与哺乳母亲一起，而常氧组中的幼崽从7-17号PND保持常氧。在PND 9,13（14）和17 [3]上对幼崽进行人道安乐死。
