[体外研究]

京尼平以时间和剂量依赖性方式刺激葡萄糖摄取。在2小时时达到最大效果,浓度为10μM。在肌管中,京尼平促进葡萄糖转运蛋白4(GLUT4)易位至细胞表面,这增加了胰岛素受体底物-1(IRS-1),AKT和GSK3β的磷酸化。同时,京尼平增加ATP水平,关闭KATP通道,然后增加C2C12肌管细胞质中钙的浓度。可以通过PI3-K抑制剂渥曼青霉素和钙螯合剂EGTA阻断京尼平刺激的葡萄糖摄取。此外,京尼平增加了C2C12肌管中活性氧和ATP的水平[1]。京尼平增加线粒体膜电位,然后增加ATP水平并关闭KATP通道,从而刺激胰腺β细胞中的胰岛素分泌。京尼平激活葡萄糖激发的POMC神经元[2]。细胞色素c含量在京尼平处理的FaO细胞的胞质溶胶中显着增加。半胱天冬酶-3和半胱天冬酶-7的激活最终导致肝细胞因子诱导的肝细胞瘤细胞凋亡过程。用200μM京尼平处理的Hep3B细胞中ROS水平显着增加[3]。

[激酶实验]

简言之，将肽底物N-乙酰基-Asp-Glu-Val-Asp-ρ-硝基苯胺（Ac-DEVD-ρNA）加入到测定缓冲液（50mM HEPES，pH 7.4,100mM NaCl，0.1％）中的细胞裂解液中。 CHAPS，10mM二硫苏糖醇，1mM EDTA，10％甘油）并在37℃下孵育。在405nm处监测底物的裂解。
