体外研究 ARCC-4诱导AR扩增前列腺癌细胞凋亡并抑制其增殖[1]。ARCC-4增强AR和VHL之间的蛋白质-蛋白质相互作用,从而促进三聚体复合体的结合[1]。ARCC-4(0.1-10000 nM;20小时)在D50为5 nM、Dmax大于95%的条件下对AR有较强的降解作用[1]。ARCC-4(100 nM;12小时)显示前列腺癌细胞中AR几乎完全降解(>98%)。ARCC-4通过蛋白酶体选择性降解AR,但不抑制PR-A或PR-B[1]。ARCC-4显示了抗临床相关AR突变的有效性[1]。尽管雄激素水平升高,ARCC-4仍保持活性[1]。Western Blot分析[1]细胞株:VCaP细胞浓度:0.1nm,1nm,10nm,50nm,100nm,0.5μM,1μM,10μM培养时间:20 h,结果:VCaP细胞有明显降解作用
