| 体外研究 | 与D2、D4和D5(pKi=8.5-8.0)相比,罗替戈汀对D3受体(pKi=9.2)具有10倍的选择性,与D1受体(pKi=7.2)相比具有100倍的选择性。在功能研究中,罗替戈汀在所有多巴胺受体上表现为完全激动剂,但值得注意的是,刺激D1受体的效力与刺激D2和D3受体的效力相似(pEC50?分别为:9.0、9.4-8.6、9.7)[1]。在原代中脑细胞培养中,罗替戈廷(10μM)可将THir神经元的数量减少40%。罗替戈汀(0.01μM)轻微保护多巴胺能神经元对抗MPP+?毒性,显著保护多巴胺能神经元免受鱼藤酮诱导的细胞死亡,并显著抑制鱼藤酮产生ROS[4]。 |
