[体外研究]

SB-3CT已在严重创伤性脑损伤(TBI)的动物模型中显示出功效。 SB-3CT抑制MMP-9的抑制常数Ki为400±15 nM [1]。通过SB-3CT抑制PC3肿瘤生长也可能是肿瘤细胞自身和/或破骨细胞在骨组织内细胞外基质降解减少的直接后果。实际上,SB-3CT治疗与溶骨性反应降低有关,表明SB-3CT有助于保持骨完整性[2]。

[体内研究]

SB-3CT后局部缺血治疗可显着减少脑损伤,并使梗塞体积减少至总半球的约20％。 SB-3CT治疗改善神经行为结果,特别是运动和感觉功能[3]。

[细胞实验]

将PC3细胞接种在完全培养基中的35-mm培养皿（5×104细胞/培养皿）中。第二天，用在1％DMSO中补充有1％DMSO单独（载体）或SB-3CT（终浓度0.1-50μM）的完全培养基替换培养基。在不同时间，用胰蛋白酶收获细胞并计数[2]。

[动物实验]

将成年雄性C57Bl / 6J小鼠，6-9周龄，体重20-28g分成四组：载体治疗组和SB-3CT治疗组，在栓塞MCA闭塞后一天或七天治疗。将SB-3CT（12.5mg / mL）新鲜溶解于25％DMSO / 65％PEG-200/10％水中，并通过具有0.2μm，13mm直径的无菌疏水性PTFE膜的Acrodisc注射器过滤器过滤。在栓塞缺血后2小时腹膜内注射2μL/克体重的该溶液（相当于25mg / kg），然后在4小时时另外注射一剂。在重复剂量治疗条件下，相同剂量的SB-3CT在栓塞性缺血后2小时和4小时腹膜内给药，然后在缺血后第1天至第6天每天一次。早期工作表明腹腔注射SB-3CT不能改变平均动脉血压，pH，PCO2和PO2。
