[体外研究]

TPCA-1显着抑制NPECs中的促炎细胞因子IL-8和GRO-α[1]。 TPCA-1抑制NFκB活性以及胶质瘤细胞增殖和IL8表达,并且在2.5和5μM的浓度下,它阻断TNF诱导的p65易位到U87细胞的细胞核中[2]。 TPCA-1抑制脂多糖诱导的人单核细胞产生TNF-α,IL-6和IL-8,IC50 = 170-320nM [3]。

[体内研究]

TPCA-1(3,10或20mg/kg,ip)导致小鼠胶原诱导的关节炎(CIA)严重程度的剂量依赖性降低。体内TPCA-1(10mg/kg,ip,bid)导致体外胶原诱导的T细胞增殖显着降低[3]。

[激酶实验]

重组人IKK-2（残基1-756）在杆状病毒中表达为N末端GST标记的融合蛋白，并且使用时间分辨荧光共振能量转移测定法评估其活性。简言之，将在测定缓冲液（50mM HEPES，10mM MgCl 2,1mM CHAPS，pH 7.4，含1mM DTT和0.01％w / v BSA）中稀释的IKK-2（最终5nM）加入含有各种浓度的孔中。化合物或二甲基亚砜（DMSO）载体（最终3％）。通过添加GST-IκBα底物（最终25nM）/ ATP（终浓度1μM），以30μL的总体积引发反应。将反应在室温下温育30分钟，然后通过加入15μL50mMEDTA终止反应。在含有抗磷酸丝氨酸-IκBα-32/36单克隆抗体12C2的缓冲液（100mM HEPES，pH7.4,150mM NaCl和0.1％w / v BSA）中的检测试剂（15μL），用W-1024铕螯合物标记，和加入别藻蓝蛋白标记的抗GST抗体，并将反应在室温下进一步温育60分钟。使用Packard Discovery平板读数器测量GST-IκBα的磷酸化程度，作为特定665-nm能量转移信号与参考铕620-nm信号的比率。

[细胞实验]

将来自储备溶液（10mg / mL）的10μL3-（4,5-二甲基噻唑-2-基）-2,5-二苯基四唑溴化物（MTT）加入到含有神经胶质瘤细胞的96孔板的每个孔中并孵育在37°C下保持2-4小时。通过在0.01N HCl中加入100μL的10％十二烷基硫酸钠（SDS）溶解氧化的MTT，并将板在37℃下在加湿室中温育4小时。在Bio-Rad平板读数器上读取570nm的平板。

[动物实验]

雄性DBA / 1 OlaHsd小鼠的年龄为6至8周。在第0天，10至12周龄雄性DBA / 1小鼠在尾巴底部皮内免疫，总共100μL含有200μg牛II型胶原和250μg结核分枝杆菌的完全弗氏佐剂。 H37Ra株。在第21天，用含有200μg牛II型胶原的100μLPBS皮内加强小鼠。在所有研究中，TPCA-1在由0.9％DMSO，7％二甲基乙酰乙酰胺（DMA）和10％Cremophor El组成的载体中施用。依那西普（Enbrel）在PBS中给药。当比较TPCA-1和依那西普的作用时，两个治疗组以及它们的相关媒介物处理的对照组都包括在同一研究中。对于预防性研究，单独使用载体或载体中的TPCA-1，从第1天开始腹膜内给药。从第1天开始每隔一天腹膜内注射PBS或PBS中的依那西普。两种载体治疗均表现出疾病的发生率。对照组（PBS-和DMSO / DMA / Cremophor-处理的小鼠）是100％。对于治疗研究，一旦动物连续2天表现出“1”或更高的临床评分，就开始给予TPCA-1，依那西普或它们各自的载体（第1天）。使用千分尺测量小鼠每天对疾病的临床症状进行评分以测量爪厚度。
