果酸(甲基原果胶)是一种聚半乳糖醛酸,可诱导垂体瘤细胞凋亡和坏死。果酸可用于癌症和自身免疫疾病的研究[2][3]。

[体外研究]

果酸(2.5-100μg/mL,30分钟)刺激GH3/B6细胞中催乳素(PRL)的释放,而不影响细胞的活性[1]。通过MTT分析和AO/EB染色检测,果酸(100μg/mL-5 mg/mL,6-48 h)增加GH3/B6细胞的细胞死亡和DNA损伤[2]。果酸(100μg/mL-1 mg/mL,24小时)以剂量依赖性方式诱导GH3/B6细胞凋亡[2]。果酸(2.5-5 mg/mL,24小时)诱导GH3/B6细胞坏死,PI染色证实[2]。细胞周期分析[2]细胞株:GH3/B6细胞浓度:1 mg/mL孵育时间:24小时结果:诱导亚G1事件,DNA断裂,这与凋亡细胞的数量相关。

[体内研究]

果酸(25和100 mg/kg,口服灌胃)增加溃疡性结肠炎(UC)小鼠的结肠长度,降低疾病活动指数、组织病理学评分和促炎细胞因子水平[3]。果酸(6.25和12.5 mg/kg,静脉注射)可挽救UC小鼠结肠长度的缩短[3]。动物模型:溃疡性结肠炎(UC)小鼠[3]剂量:25100 mg/kg给药:口服灌胃结果:结肠最长,DAI最低,组织病理学评分最低。降低UC小鼠结肠组织中的促炎细胞因子。动物模型:溃疡性结肠炎(UC)小鼠[3]剂量:6.25,12.5 mg/kg给药:静脉注射结果:脾脏和胸腺指数降低。恢复UC小鼠脾脏和固有层的Th17/Treg平衡。改善肠道菌群组成。
