5 μM (MDA-MB-231)

体外研究 甲磺酸苯托品(0.1-10μM;72小时)处理抑制MDA-MB-231细胞的生长,IC50约为5μM。在MDA-MB-231细胞和4T1-luc2细胞中,甲磺酸苯托品以剂量依赖性甘露醇显著减少乳腺球的大小和数量[1]。甲磺酸苯托品通过乙酰胆碱受体、多巴胺转运体/受体和/或组胺受体抑制肿瘤干细胞的功能[1]。甲磺酸苯托品通过M1和M3毒蕈碱受体诱导少突胶质细胞分化,并增强髓鞘再生[1]。细胞活力测定[1]细胞系:MDA-MB-231浓度:0,0.1,0.625,1.25,2.5,5,10μM培养时间:72小时结果:IC50~5.0μM抑制MDA-MB-231细胞生长。细胞活力测定[1]细胞系:MDA-MB-231浓度:0、1、2、5μM孵育时间:4-6天结果:体外以剂量依赖性方式抑制乳腺球的形成和基底细胞干细胞的自我更新能力。
体内研究 甲磺酸苯托品(5μM,4周)在4T1小鼠模型中抑制肿瘤起始电位[1]。动物模型:携带4T1乳腺肿瘤的Balb/c小鼠[1]剂量:1.5mg/kg给药:注射;3周结果:肿瘤体积和重量显著减小,体重无变化。
