Dihydrocoumarin 是在 Melilotus officinalis 中发现的一种化合物。Dihydrocoumarin 是酵母 Sir2p 抑制剂。 Dihydrocoumarin 还抑制人 SIRT1 和 SIRT2,IC 50 分别为 208 μM 和 295 μM[1]。

[体外研究]

在体外酶促测定中,二氢香豆素诱导SIRT1的浓度依赖性抑制(IC50为208μM)。即使在微摩尔剂量下也观察到SIRT1脱乙酰酶活性的降低(分别为1.6μM和8μM时的85±5.8和73±13.7％活性)。微管SIRT2脱乙酰酶也被抑制,具有相似的剂量依赖性(IC50为295μM)[1]。在24小时暴露后,二氢香豆素(1-5mM)以剂量依赖性方式增加TK6细胞系中的细胞毒性。二氢香豆素(1-5mM)在6小时时间点在TK6细胞系中以剂量依赖性方式增加细胞凋亡。 5mM剂量的双氢香豆素在TK6细胞系的6小时时间点增加细胞凋亡[1]。在24小时暴露期后,二氢香豆素(1-5mM)在TK6细胞系中以剂量依赖性方式增加p53赖氨酸373和382乙酰化[1]。
