[体外研究]

在WR-1065二盐酸盐(WR-1065)浓度依赖性方式中,DNA结合活性增加。用1mM WR-1065二盐酸盐处理24小时的细胞显示所有分析的p53诱导的基因在WR-1065二盐酸盐处理后以p53依赖性方式反式激活。值得注意的是,用WR-1065二盐酸盐处理导致AP-1驱动的荧光素酶表达增加3倍,当这种报告基因被NF-κB驱动时增加5倍,当这些值被归一化到水平时共转染的β-半乳糖苷酶基因[2]。

[体内研究]

结果显示,与6-OHDA损伤的大鼠相比,wR-1065二盐酸盐(WR-1065)减弱了6-OHDA诱导的强直性昏厥的严重程度(P <0.001)。还观察到WR-1065二盐酸盐以剂量依赖性方式改善僵住症(P <0.001)。在施用6-OHDA之前,用三种不同剂量的WR-1065二盐酸盐(20,40和80μg/2μL/大鼠)预处理3天,显着(P <0.001)提高SOD活性并将其恢复至正常范围与6 -OHDA损伤的大鼠[3]。

[激酶实验]

对于Western分析，用1mM WR-1065二盐酸盐（WR-1065）处理细胞24小时，收获细胞的亚汇合培养物并在补充有蛋白酶抑制剂的RIPA缓冲液中裂解。蛋白质浓度通过与洗涤剂相容的测定来确定。阻断Western印迹并在PBS / 0.2％Tween 20/5％脱脂奶粉中的抗体中温育。将印迹与1μg/ mL抗体在室温下孵育1小时，然后在PBS / 0.2％吐温20中洗涤并在过氧化物酶缀合的二抗中孵育并进行化学发光检测[2]。

[细胞实验]

为了测试紫杉醇在WR-1065二盐酸盐（WR-1065）存在或不存在下对细胞生长的影响，将细胞以20％汇合接种于96孔组织培养皿中并使其附着并恢复至少24小时。。然后将紫杉醇单独或与用1mM WR-1065二盐酸盐预处理60分钟组合的不同组合加入每个孔中，并将板再温育48小时或72小时。通过染色确定存活细胞的数量。紫杉醇和/或WR-1065二盐酸盐杀死细胞的百分比计算为与对照细胞相比磺酰罗丹明B结合的降低百分比[2]。

[动物实验]

将72只大鼠随机分成9个相等的组：1）对照组不接受注射，并且作为完整动物在整个实验期间不进行处理; 2）假手术组仅接受外科手术; 3）载体（盐水）处理组接受2μL盐水（SNc内）; 4）损伤组接受6-羟基多巴胺; 5）载体+ 6OHDA组在6-OHDA注射前每天一次（2μL/大鼠）接受盐水作为载体; 6至8）在6-OHDA注射前3天，用SNc内注射WR-1065二盐酸盐（WR-1065）（20,40和80μg/2μL/大鼠）预处理这些组中的大鼠; 9）未损伤的动物接受SNc内注射WR-1065二盐酸盐（80μg/2μL/大鼠）3天[3]。
