利美尼啶半富马酸盐用途

Rilmenidine hemifumarate 是一种新型的抗高血压药物和口服活性选择性 I1 咪唑啉受体 (I1 imidazoline receptor) 激动剂。Rilmenidine hemifumarate 也是一种 α2 肾上腺素受体激动剂。Rilmenidine phosphate 诱导自噬 (autophagy)。Rilmenidine hemifumarate 可通过减少交感神经过度活跃而发挥中枢作用,并通过抑制 Na+/H+ 反向转运而在肾脏中发挥作用。Rilmenidine hemifumarate可调节白血病细胞增殖,刺激促凋亡蛋白 Bax,从而诱导人白血病 K562 细胞线粒体通路的紊乱和凋亡。

[体外研究]

利美定的降压疗效与利尿剂、β受体阻滞剂、钙通道阻滞剂和血管紧张素转换酶(ACE)抑制剂相当[1]。Rilmenidine(25-100μM；24小时)抑制K562细胞增殖[2]。细胞活力测定[2]细胞系：K562细胞浓度：25、50、100μM孵育时间：24小时结果：剂量依赖性抑制K562集落形成。

[体内研究]

利美尼定治疗的N171-82Q小鼠(腹腔注射；每周4次)在12至22周龄时显示出前肢握力和所有肢体握力的显著改善[3]。利美定降低突变亨廷顿蛋白的水平[3]。
