[体外研究]

与对照相比,盐酸克伦特罗增加大鼠原代脂肪细胞的脂解作用。在含有0.1或1μM盐酸克伦特罗的培养物中,游离甘油释放到培养基中的对照值分别为对照值的158%和190%[1]。

[体内研究]

盐酸克伦特罗已被证明可以降低动物体内脂肪,并可诱导小鼠脂肪组织细胞凋亡[1]。在红色和白色肌肉中,盐酸克伦特罗诱导线粒体含量降低,参与脂肪酸转运氧化的蛋白质,葡萄糖转运,乳酸转运,单羧酸转运蛋白和丙酮酸氧化。克伦特罗引起的这些广泛的代谢变化与PGC-1α的减少和RIP140的增加有关[2]。对大鼠重复施用中枢作用的β肾上腺素能受体激动剂克伦特罗降低了异丙肾上腺素增加小脑切片中环AMP(cAMP)浓度的能力。这种对异丙肾上腺素的反应性降低伴随着β肾上腺素受体密度的显着降低[3]。在正常比目鱼肌中,盐酸克仑特罗处理刺激蛋白质合成,抑制Ca2 +依赖性蛋白水解,并增加钙蛋白酶抑制蛋白的水平。另一方面,给予DEN大鼠的盐酸克伦特罗改善了肌肉质量的损失,提高了蛋白质合成的速度,减弱了蛋白酶体和溶酶体蛋白水解的过度活化,并抑制了溶酶体蛋白酶组织蛋白酶L和atrogin-1的转录。 MAFbx和MuRF1 [4]。

[细胞实验]

进行MTS测定以确定培养物中存活细胞的数量。用0(仅载体),0.1或1μM盐酸克伦特罗处理细胞6,12,24或48小时。在读板器中在490nm处测量吸光度,确定甲amount浓度,其与培养物中活细胞的数量成比例[1]。

[动物实验]

大鼠:将大鼠随机分成两个治疗组。对照组随意提供标准食物和水3周,而克伦特罗组提供标准食物,并通过饮用水(30mg盐酸克伦特罗/ L)给予克伦特罗治疗3周。每周记录两次体重和耗水量。 3周后,将动物麻醉并切除后肢肌肉[2]。
