[体外研究]

异环磷酰胺是一种烷化化学治疗剂,具有抗多种肿瘤的作用[1]。异环磷酰胺被细胞色素P450家族激活。异环磷酰胺(0-5 mM)抑制表达CYP2B1的C8III-1细胞的活力,而仅在高浓度(5 mM)时对表达非CYP2B1的CrFK细胞具有细胞毒性[2]。 CYP BM3突变体激活异环磷酰胺,异环磷酰胺对人U2OS细胞具有抑制活性[3]。

[体内研究]

交配前的异环磷酰胺(50mg/kg,ip)处理增加了植入后损失的百分比并且在怀孕大鼠中吸收胎儿。异环磷酰胺(50mg/kg,ip)也降低了怀孕大鼠血清中的孕酮。然而,异环磷酰胺与25mg/kg对照大鼠没有明显差异。异环磷酰胺(25,50 mg/kg,ip)诱导胎盘和产前大鼠的细胞凋亡和组织学变化,最敏感的胎儿器官是脑,肝和肾[1]。

[细胞实验]

在3cm培养皿中,将4×104个细胞接种在2mL培养基中。第二天,加入异环磷酰胺至终浓度为0至5 mM。再过6天后,除去培养基,用PBS洗涤细胞,染色或计数[2]。

[动物实验]

大鼠[1]在交配之前,将雌性大鼠分成四组,每组八只:第1组,未治疗的阴性对照;第2组,大鼠腹腔注射1 mL 0.9%NaCl;第3组,大鼠腹腔注射25 mg / kg异环磷酰胺;第4组,大鼠腹腔注射50 mg / kg异环磷酰胺。在每天一次注射异环磷酰胺连续5天后,将三只雌性与一只未经治疗的雄性一起放置在笼子中长达1周。每天检查阴道涂片以确定怀孕。如果检测到精子,则在交配后的第一个24小时期间被指定为怀孕第一天。怀孕的雌性单独饲养,每天观察有毒性和流产的迹象。在妊娠第18天通过断头处死所有怀孕动物。在非肝素化试管中收集心脏血液(2.5-3mL /大鼠),以3,000×g离心30分钟,倾析血清并储存在-70℃直至用于激素测定。收集血液后,取出子宫和两个卵巢,用盐水溶液洗涤,目测计数妊娠黄体,并计算每个子宫角的植入部位,再吸收部位和活胎的数量。将每个胎儿从其脐带中取出,称重并测量冠臀(CR)长度。检查胎儿的外部畸形并记录胎盘重量。对照组和治疗组的胎儿和胎盘用10%中性缓冲福尔马林固定,用于免疫组化和组织学检查[1]。
