[体外研究]

GPR35作为犬尿氨酸途径中间犬尿喹啉酸的受体起作用。在G qi/o嵌合G蛋白存在下,犬尿喹啉酸以GPR35依赖性方式引发钙动员和肌醇磷酸生成。犬尿喹啉酸在表达GPR35的细胞中刺激[35S]鸟苷5'-O-(3-硫代三磷酸)结合,这种作用被百日咳毒素处理消除。犬尿喹啉酸还诱导GPR35的内化[1]。使用毫摩尔范围内的化合物浓度发现了KYNA的神经抑制特性及其神经保护和抗惊厥作用。这,以及KYNA对负责这些作用的三种离子型谷氨酸受体中的每一种的低亲和力[NMDA,α-氨基-3-羟基-5-甲基-4-异恶唑丙酸(AMPA)和红藻氨酸],认识到哺乳动物大脑中的KYNA浓度在亚微摩尔范围内,表明其他受体可能作为内源性犬尿喹啉酸的靶标。犬尿喹啉酸具有较浅的抑制曲线且非竞争性,可拮抗培养的海马神经元上的α7nAChRs,IC50在低微摩尔范围内[2]。

[体内研究]

犬尿喹啉酸影响小鼠外周血中白细胞的活性,尽管最低的(2.5mg/L)具有最显着的影响,而最高的(250mg/L)产生最弱的效果。在给动物施用酸7和28天后,最低的犬尿喹啉酸剂量刺激T淋巴细胞的增殖反应(p <0.05)[3]。

[激酶实验]

在收获前，用或不用百日咳毒素（100ng / mL）预处理CHO-GPR35稳定细胞16小时。将细胞重新悬浮并在10mM Tris-HCl（pH 7.4），1mM EDTA中匀浆，然后在4℃下以1000×g离心10分钟以除去细胞核和细胞碎片。通过将上清液以38,000×g旋转30分钟收集膜级分，并重悬于20mM HEPES（pH 7.5）和5mM MgCl 2中。将25μg膜在室温下在含有3μMGDP和0.1nM [35S]GTPγS的测定缓冲液（20mM HEPES，5m MMgCl2,0.1％牛血清白蛋白（pH7.5））中孵育1小时，不存在或存在犬尿喹啉酸。通过GF / B过滤器真空过滤终止反应，并在液体闪烁计数器上定量保留的放射活性[1]。

[动物实验]

小鼠：实验在160只雄性BALB / c小鼠上进行，年龄10-12周，体重22-26g。在控制温度（20±1℃）下将动物维持在12小时光照/黑暗循环中，并在整个实验中随意提供啮齿动物食物和水。将小鼠随机分成四个相等的组：对照组（0）不接受犬尿喹啉酸，三个实验组在饮用水中以2.5,25或250mg / L的浓度给予犬尿喹啉酸溶液。在给予犬尿喹啉酸溶液连续3天，7天，14天和28天后，处死每组10个个体。通过吸入Aerrane麻醉动物，并通过心脏穿刺收集它们的血液。从每组的5个个体收集的血液用于MTT测定，并且从接下来的5个用于流式细胞术[3]。
