[体外研究]

Withaferin A具有抗炎活性,通过硫代烷基化敏感的氧化还原机制阻止TNF诱导的Ik-B激酶β活化,从而有效抑制NF-κB活化[1]。 Withaferin A也具有抗癌活性。 Withaferin A靶向IF蛋白波形蛋白,导致牛主动脉内皮细胞(BAECs)波形蛋白丝聚集3μM,并诱导10μM内皮细胞波形蛋白片段化[2]。单独使用Withaferin A(0.5,1.5μM)或与顺铂(CIS)联合剂量依赖性地降低ALDH1阳性癌症干细胞(CSC)的致瘤潜力[3]。

[体内研究]

Withaferin A(2 mg/kg,ip)通过波形蛋白在小鼠中显示出有效的血管生成抑制活性[2]。 Withaferin A(2 mg/kg)联合顺铂(CIS)调节ALDH1标志物的表达,下调从小鼠收集的肿瘤中securin的表达[3]。

[细胞实验]

将卵巢上皮癌细胞系A2780维持在补充有胰岛素（5μg/ mL），青霉素/链霉素（分别为100IU / mL和100μg/ mL）和来自Hyclone的10％胎牛血清（FBS）的RPMI1640培养基中。在DMSO中制备Withaferin A，顺铂（CIS）和其他试剂。顺铂每次都是新鲜制备的[3]。

[动物实验]

在测定中使用小鼠[2]波形蛋白纯合缺陷小鼠（Vim - / - ）和129 / Svev背景中波形蛋白 - 杂合缺陷（Vim +/-）的小鼠。简言之，通过腹膜内（ip）注射氯胺酮和甲苯噻嗪麻醉4至6周龄的小鼠。通过应用proparacain滴眼液局部麻醉角膜，并施加1μL稀释的0.15M氢氧化钠1分钟。角膜立即在盐水溶液中充分洗涤，通过刮擦轻轻除去角膜和角膜缘上皮。用阿托品滴眼液局部处理角膜，并用妥布霉素和红霉素抗生素眼膏覆盖。在角膜损伤恢复后，将Withaferin A或12-D WS（2mg / kg溶于DMSO中）或载体（DMSO）腹膜内注射到相应的药物或对照组小鼠中，随后每天注射10天。。小鼠被人道地杀死并且眼睛去核。解剖眼睛的前半部分并准备角膜按钮。将角膜组织在100％丙酮中固定20分钟，在PBS中洗涤1小时，并在4℃下在1％BSA-PBS中封闭18小时。角膜整体染色通过将组织在FITC缀合的大鼠抗小鼠CD31抗体（在1％BSA-PBS中1：333稀释）中孵育12小时，在4℃下在1％BSA中洗掉24小时来进行。 PBS，用盖玻片固定在载玻片上。荧光染色在显微镜上可视化，并通过将数字图像导入NIH ImageJ来量化[2]。
