索法酮用途

本品用来生产索法酮药品。 索法酮成品药能扩张胃粘膜血管,增加胃粘膜血流量和胃组织耗氧量,促进胃粘膜修复。还能抑制前列腺素分解酶,从而增加胃内前列腺素含量。索法酮成品药主要通过增强防御因子而对消化性溃疡发挥良好的作用。

Sofalcone 是临床使用的胃溃疡剂,可以诱导血红素加氧酶-1 (HO-1) 在胃肠上皮中的表达。

[体外研究]

与滋养细胞和HUVECs中的对照组相比,Sofalcone(50μmol/ L)显着增加HO-1 mRNA表达(两者均P <0.05)。蛋白质印迹分析表明,Sofalcone有效增加处理24小时的原代HUVEC中的HO-1蛋白表达。蛋白质印迹分析还显示,使用Sofalcone处理(50μmol/ L)6小时,HUVEC核部分中Nrf2的量显着增加。在原代HUVEC中,使用Sofalcone处理24小时,NQO1,TXN和GCLC的表达以剂量依赖性方式增加。 Sofalcone显着降低培养基中sFlt-1的浓度,剂量依赖性地降低THP-1单核细胞对HUVECs的依从性[1]。

[细胞实验]

将原代HUVEC以每孔60 000个细胞接种,并在37℃下孵育14至16小时以允许形成管。然后在TNF-α(10ng / mL)单独存在下,用TNF-α(10ng / mL)和Sofalcone50μM或对照培养基培养细胞8至12小时。评估管形成,并使用显微镜以×4放大率捕获图像[1]。
