[体外研究]

4-甲基伞形酮(4-MU)影响血管生成的几个关键步骤,包括内皮细胞增殖,粘附,管形成和细胞外基质重塑。对于HMEC和RF-24内皮细胞,增殖测定中的半数最大抑制浓度(IC 50)值分别为0.65±0.04和0.37±0.03mM。 4-甲基伞形酮(2mM)处理24小时诱导13%的HMEC和5%的RF-24细胞凋亡。在用1mM 4-甲基伞形酮处理24小时后,粘附的内皮细胞的数量减少> 20%。对于HMEC和RF-24,管形成测定中的最小抑制浓度分别为2和0.5mM 4-甲基伞形酮。在两种测试的内皮细胞系中,基质金属蛋白酶-2表达在4-甲基伞形酮治疗后差异性改变[1]。

[细胞实验]

使用96孔微量培养板中的MTT染料还原测定。该测定依赖于活细胞的线粒体脱氢酶对蓝色甲product产物的MTT降低,其可以通过分光光度法测量。将内皮细胞(总体积为100μL完全培养基中的2.5×103细胞)在每个孔中与4-甲基伞形酮(4-MU)(0,0.5,1,1.5和2mM)的连续稀释液一起温育。在黑暗中(37℃,5%CO 2在潮湿气氛中)孵育3天后,向每个孔中加入10μLMTT(在PBS中5mg / mL),并将板再孵育4小时。 (37℃)。将甲crystals溶解在150μL的0.04N HCl-2丙醇中,并在550nm下用分光光度法测量样品。所有测定均以一式四份进行,并且进行至少三次独立实验。 IC50值计算为产生50%细胞存活率的化合物浓度,将对照获得的值作为100%[1]。
