[体外研究]

Nociceptin(1μg/ mL)显着阻止LPS(10 ng/mL)刺激的细胞迁移,而单独添加时无效。 Nociceptin(1 nM-10μM)引发LPS介导的细胞迁移的浓度依赖性阻断,在1和10μM时具有最大效应。 Nociceptin抵消LPS诱导的IL-1βmRNA水平升高。 Nociceptin(1μM)和NNC 55-0396在U87细胞中诱导凋亡细胞死亡。 Nociceptin(1μM)通过β-抑制蛋白2抵消LPS诱导的U87细胞[Ca2 +] i增加.Nociceptin抵消LPS诱导的U87细胞中PKC和ERK的磷酸化。 Nociceptin抑制LPS介导的NF-kB和AP-1报告基因的转录激活[1]。

[细胞实验]

在测定中进行细胞增殖测定。将U87细胞接种在12孔板上，并在含有10％胎牛血清的细胞培养基中维持24小时。在处理结束前5小时，将[甲基-3H]胸苷（50nM终浓度）加入无血清细胞培养基中，并将板在37℃温育。此后，除去培养基并用PBS洗涤细胞两次。向每个孔中加入200μLPBS，刮下细胞并在4℃下以13,000g离心3分钟;然后弃去上清液，将沉淀重悬于500μL冷三氯乙酸（10％w / v）中，在冰上孵育20分钟，并在4℃下以13,000g离心3分钟。然后弃去获得的上清液，将沉淀物悬浮在500μL冷甲醇中，并在4℃下在3分钟离心13,000g。之后，将沉淀物悬浮在200μL1NNaOH中并在55℃下加热10分钟。然后用200μL1NHCl中和样品，并在计数小瓶中用4mL Filter Count闪烁液孵育350μL标记的DNA。将小瓶涡旋并在室温下温育过夜，并通过液体闪烁光谱法测定放射性。
