[体外研究]

聚维酮碘(PVP-1)的MIC值分别为31.25mg/L和7.82mg/L.用聚乙烯吡咯烷酮碘(PVP-I)以1:32的稀释度处理细胞导致所有测试细胞系的细胞活力急剧下降90-95%[1]。

[体内研究]

Dp +聚维酮碘(PVP-I)组从第13天到实验期结束的平均得分第二高。与对照组相比,Dp +聚维酮碘和Dp + Et-OH基团也显示嗜酸性粒细胞计数显着增加(分别为p <0.05和p <0.001)。然而,嗜酸性粒细胞计数在Dp +聚维酮碘(PVP-1),Dp + CHG和Dp +载体组之间没有显着差异[2]。

[细胞实验]

将每种细菌分离物用磷酸盐缓冲盐水(PBS,pH7.2)洗涤两次,在1932xg,20℃下离心10分钟,并悬浮在3mL营养肉汤中,调节至浊度当量为0.5McFarland标准。将细菌悬浮液用MHB以1:100稀释至106个菌落形成单位(cfu)/ mL的最终接种物。对于每种细菌菌株,用最终的细菌接种物(每孔50μL)和50μL的每种连续稀释的ILαD填充两排96孔微量滴定板。对聚维酮碘(PVP-I)系列稀释液重复该过程[1]。

[动物实验]

将小鼠分成如下6组:1)盐水+载体(对照组),2)Dp +载体,3)Dp + BZK,4)Dp +聚维酮碘(PVP-1),5)Dp + Et- OH,和6)Dp + CHG。实验组中的动物通过在麻醉下每周2至3天(总共8次)皮下注射溶解于右耳腹侧的10μL生理盐水中的5μgDp暴露于过敏原。 %氟烷。对照组中的动物未致敏,在右耳腹侧接受10μL生理盐水的皮下注射。动物接受抗菌剂暴露于过敏原并用0.2%(w / v)苯扎氯铵(Dp + BZK),10%(w / v)聚维酮碘(Dp + PVP-I),80%处理(v / v)乙醇(Dp + Et-OH)或0.5%(v / v)葡萄糖酸氯己定(Dp + CHG)。这些试剂在实验期间共施用15次。将BZK,聚维酮碘(PVP-I),Et-OH和CHG溶解在25μL注射用水中,并使用具有细塑料尖端的微量移液管轻轻地施加到右耳的背侧。 Dp +载体和对照组中的动物接受25μL注射用水。在实验的最后一天(第18天)处死所有动物[2]。
