[体外研究]

Berbamine是一种新型NF-κB抑制剂,可抑制人骨髓瘤细胞的生长并诱导细胞凋亡。小檗胺处理导致A20的表达增加,IKKα,p-IκBα的下调,并且随后抑制p65核定位。结果,NF-κB下游靶标如细胞周期蛋白D1,Bcl-xL,Bid和存活蛋白被下调。为了确定小檗胺是否对骨髓瘤细胞具有生长抑制作用,分别用不同浓度的小檗胺处理KM3细胞24,48和72小时,然后通过MTT测定评估细胞活力。 Berbamine以剂量和时间依赖性方式抑制KM3细胞的生长,并且对于24,48和72小时的处理,IC50值分别为8.17μg/ mL,5.09μg/ mL和3.84μg/ mL。相比之下,Berbamine对正常造血细胞的IC50值在48小时时为185.20μg/ mL [1]。

[体内研究]

小檗胺(BBM)是从中草药小檗中分离的天然双苄基异喹啉产品,已被用于治疗炎症和其他疾病。小檗胺的抗肿瘤作用是在异种移植物动物模型上测定的。通过皮下注射将两种肝癌细胞系Huh7(上皮)和SK-Hep-1(间充质样)接种到NOD/SCID小鼠中。口服小檗胺处理在很大程度上抑制了Huh7异种移植肿瘤的生长,并且基于肿瘤重量导致肿瘤减少70％。 NOD/SCID小鼠中SK-Hep-1细胞的生长对小檗胺的敏感性低于Huh7。 SK-Hep-1异种移植物的生长受到显着抑制,肿瘤重量减少超过50％[2]。

[细胞实验]

通过MTT测定法测量小檗胺对KM3细胞生长的抑制作用。简言之，将KM3细胞（每孔8×103）与浓度增加的小檗胺（1-32μg/ mL）一起孵育24,48或72小时，然后用20μL5mg / mL MTT进行脉冲处理4小时h，然后加入200μLDMSO以溶解甲crystals晶体。在570nm处测量活细胞中的染料吸光度，然后计算50％的抑制浓度（IC50）[1]。

[动物实验]

将溶解在PBS中的50％基质胶中的小鼠[2] 5×106 Huh7细胞接种在NOD / SCID小鼠中。对于没有Matrigel的每个异种移植物应用5×106个SK-Hep-1细胞。在肿瘤达到直径2mm的大小后，以每天两次的方案对小鼠口服100mg / kg的小檗胺，连续5天。退出2天后，方案重复一次[2]。
