[体外研究]

青霉素G钠盐-TEM-1体系的紫外-可见(UV-Vis)吸收光谱与青霉素G钠盐和TEM-1β-内酰胺酶的紫外-可见(UV-Vis)吸收光谱明显不同,表明青霉素G钠之间形成新的复合物。盐和TEM-1β-内酰胺酶。随着青霉素G钠盐浓度的增加,TEM-1β-内酰胺酶的紫外-可见吸收增加,并发生轻微的红移,表明青霉素G钠盐与TEM-1β-内酰胺酶之间的相互作用导致微妙的构象。 TEM-1β-内酰胺酶的变化[1]。

[体内研究]

在Logistic回归模型中,对照组阳性拭子的概率比用青霉素G钠盐处理的猪高1.6倍(P <0.05)。在对照组中,每个平板具有10至99个菌落的拭子与每个平板具有零的风险相比,是用青霉素G钠盐处理的猪的2.3倍(P = 0.022)[2]。

[激酶实验]

在278K,将各种浓度的青霉素G钠盐,头孢氨苄和头孢西丁溶液加入到TEM-1β-内酰胺酶溶液(5×10-6M)中。三种抗生素的浓度从0逐渐增加到25×10-6M。在混合和相互作用2分钟后,在具有2nm狭缝的分光光度计上记录紫外 - 可见(UV-Vis)吸收光谱。使用0.02M磷酸盐缓冲液(pH7.0)作为参考[1],扫描速度为400nm / min。

[动物实验]

本研究使用具有2个重复的随机完整区组设计。每个重复包括448头猪,每支笔有16支笔和28头猪。猪也按重量分类;使得基于视觉观察的相似重量的动物在块内组合在一起。使用正式随机化过程在2个连续笔的每个块内随机分配两种处理。治疗组是对照(未给予治疗)和治疗(青霉素G钠盐)。青霉素G钠盐在2个治疗期间通过饮用水给药5天。第一个治疗期从断奶当天开始,当时猪被移入托儿所(第1天)并在第5天结束。第二个治疗期从第21天开始到第25天结束。对照组未接受治疗治疗[2]。
