[体外研究]

邻苯二甲酸二丁酯(0.001μg/ mL-1000μg/ mL)对卵泡生长和活力有害,并且以剂量特异性方式导致细胞周期和凋亡基因表达的显着失调。但MBP在体外暴露的卵泡中对邻苯二甲酸二丁酯的毒性没有作用[1]。

[体内研究]

邻苯二甲酸二丁酯(200,400或600 mg/kg /天)诱导减轻小鼠体重,减少精子发生,降低血清卵泡刺激素和睾酮水平,改变睾丸LDH,增加LPO,并降低酶抗氧化剂水平与组织病理学异常[2]。邻苯二甲酸二丁酯(6.25,12.5,25,50,100和200 mg/kg)可能对小鼠造成一些神经行为不良反应[3]。

1. 以邻苯二甲酸酐和丁醇为原料(其质量比为1:1.5),以硫酸为催化剂(用量为苯酐和丁醇质量之和的0.3%),为了防止醇的氧化,加入总投料量0.1%~0.3%的活性炭,起吸附氧气和脱色作用。加热,在常压下进行酯化。酯化反应在耐酸搪瓷釜或不锈钢釜中进行。当固体苯酐全部溶解,说明酯化反应的第一步生成邻苯二甲酸单丁酯的反应已结束,这步反应迅速而完全。然后继续加热至反应物沸腾,进行第二步生成邻苯二甲酸二丁酯的反应,液相温度150℃左右,时间4~5h。此阶段不断蒸出醇水恒沸物,经冷凝器冷凝后进入醇水分离器。丁醇返回反应釜,下层水排除。当反应温度升到170℃,冷凝液中不再有水珠,取样测定酸值,若酸值达终点则降温至70℃左右,加入5%的纯碱溶液中和,搅拌0.5h,静置1h,分去水层。粗酯再用70~80℃的热水洗涤。然后进行真空蒸馏,回收丁醇,当压力为7.9Pa,温度为150℃左右,粗酯闪点达160℃以上,脱醇完成。再用直接蒸汽吹出低沸物,经板框压滤机压滤,即得成品。

2. 精制方法:含有游离的酸、醇和邻苯二甲酸一丁酯等杂质。精制时用水洗涤除去醇和酸,再用稀的氢氧化钠溶液洗涤除去邻苯二甲酸一丁酯,然后用无水碳酸钾或硫酸钠干燥,减压下蒸馏。
