[体外研究]

在浓度>68μg/ mL之前,暴露于TDCPP不会显着影响细胞活力。在暴露于136μg/ mL TDCPP后,HCEC显示16％的细胞活力丧失。此外,TDCPP在暴露于≥272μg/ mL TDCPP后诱导活细胞急剧减少(87％)。基于细胞活力,使用非线性回归,TDCPP的LC50值为202μg/ mL。与对照相比,暴露于TDCPP的细胞表现出细胞凋亡的浓度依赖性增加。暴露于2μg/ mL TDCPP后,抗凋亡Bcl-2蛋白表达增加至1.4倍,在20μg/ mL时增加1.2倍,但与对照相比,在200μg/ mL时动态减少至0.4倍。在200μg/ mL TDCPP下,caspase-3活性增加至对照的2.1倍[1]。 TDCPP在较低浓度下抑制细胞生长(IC50为27μM),而细胞活力和毒性在较高浓度下受到影响(IC50分别为171μM和168μM)[2]。

[激酶实验]

根据制造商的指南，使用基于荧光素酶的ATP测定试剂盒，在含有0,2,20或200μg/ mL TDCPP的DMEM中生长的HCEC中测定细胞ATP含量。简而言之，暴露24小时后，用裂解缓冲液裂解HCEC。然后将裂解物在12,000g，4℃下离心5分钟。然后，将100μL上清液与100μLATP检测工作稀释液混合。通过荧光酶标仪[1]检查亮度。

[细胞实验]

为了检查TDCPP对细胞活力的影响，将HCEC以1×10 5个细胞/ mL的密度种植到96孔板（100μL/孔）中过夜。然后，将培养基更换为含有0.034,0.34,3.4,34,68,136,272或340μg/ mL TDCPP和溶剂载体（0.1％，v / v）的新鲜培养基，并培养24小时。根据制造商的说明，使用CCK-8细胞活力测定试剂盒检测细胞活力。暴露后，通过倒置显微镜观察并记录细胞形态[1]。
