[体外研究]

浓度高于0.0025%的硫酸卡那霉素对双歧双歧杆菌的生长具有显着的抑制作用,并且在孵育12小时或24小时时对其他四种益生菌没有影响。对于B.bifidum的选择性计数,MRS培养基中硫酸卡那霉素的最佳选择浓度为0.005%[3]。

[体内研究]

由卡那霉素(500mg/kg /天)引起的DCN的神经元损伤是可逆的,并且通过JNK1介导的大鼠Bcl-2途径的磷酸化,在DCN上卡那霉素的神经毒性过程中自噬被上调。在卡那霉素给药期后第1天血清BUN和Cr水平均升高。与对照组相比,在卡那霉素给药后第1,7和14天,表达LC3的神经元增加。卡那霉素治疗以时间依赖的方式导致自噬增加[1]。硫酸卡那霉素(5 mg/kg)和氨苄西林钠(10 mg/kg)分别肌肉注射(im),然后一起给予5匹小马,并且在发炎的滑液中氨苄青霉素浓度超过5 mg/mL约2.5 h注射后,硫酸卡那霉素浓度超过2 mg/mL,在小马中持续7 h [2]。

[动物实验]

六十六只雄性Sprague-Dawley大鼠(初始体重125-150g,5-6周龄)可自由饮水和定期饮食,并且在第一次治疗前允许1周适应。将动物随机分成一个对照组和七个实验组。对照组大鼠(n = 10)皮下注射等体积的载体(0.9%盐水)10天,与卡那霉素处理组相同,但不含卡那霉素。实验组(每组n = 56,8:分别给予卡那霉素后1,7,14,28,56,70和140天)通过皮下注射接受500mg硫酸卡那霉素/ kg /天,持续10天。每天监测动物体重,并相应地调整卡那霉素的注射剂量。听觉阈值由ABR测试。每只动物进行两次试验,首先在给药开始之前,然后在卡那霉素处理后的不同观察时间点进行。 ABR测量的细节在别处描述。
