尼群地平制备

1.间硝基苯甲醛和乙酰乙酸乙酯在酸催化下缩合，得2-(3-硝基苄亚基)乙酰乙酸乙酯。
2-(3-硝基苄亚基)乙酰乙酸乙酯和3-氨基丁烯酸甲酯置索氏提取器烧瓶中，加入无水乙醇，索氏提取器的虹吸管中装入滤纸包严的分子筛，水浴加热回流4h后，再放置过夜。过滤析出的结晶，用5倍量的无水乙醇重结晶，得尼群地平纯品，熔点158～159℃，收率90.8%。如果不用分子筛，收率为86.1%。

2.硝基苯甲醛和乙酰乙酸乙酯在酸催化下缩合，得2-（3-硝基苄亚基）乙酰乙酸乙醇。该物质和3-氨基丁烯酸甲酯放入索氏提取器烧瓶中，加入无水乙醇，索氏提取器的虹吸管中装入滤纸包严的分子筛，水浴加热回流、静置。过滤析出的结晶，经重结晶，得尼群地平纯品。
