尼莫地平制备

方法1:间硝基苯甲醛先和乙酰乙酸甲氧基乙酯在盐酸或浓硫酸催化下缩合,得到的产物再和3-氨基丁烯酸异丙酯在无水乙醇中加热环合,即可得尼莫地平。


方法2:间硝基苯甲醛也可先和乙酰乙酸异丙酯于室温下搅拌溶解,加入冰醋酸和哌啶,在40-50℃搅拌至固化(约6~7h)。在40-50℃静置保温4h后,加入95%乙醇,加热回流至固体溶解。冷到0-5℃,过滤析出的结晶,干燥,得2-(3-硝基苯亚甲基)乙酰乙酸异丙酯,收率90.6%。然后和3-氨基丁烯酸2-甲氧基乙酯反应,粗品用乙醇重结晶,得尼莫地平,收率84%,熔点125~126℃。


第二步也可如下进行:3.8g 2-(3-硝基苯亚甲基)乙酰乙酸异丙酯、8g乙酰乙酸2-甲氧基乙酯和6ml浓氨水在80ml乙醇中,加热回流8h。产物用石油醚-乙酸乙酯重结晶,即得尼莫地平,收率49%,熔点125℃。
