[体外研究]

辣椒素是瞬时受体电位香草酸亚型1(TRPV1)的激动剂,其在伤害性感觉神经元和一系列分泌性上皮细胞(包括唾液腺)中表达。辣椒素激活TRPV1,其通过以ERK依赖性方式调节推定的细胞间粘附分子的表达和功能来调节紧密连接(TJ)的通透性[2]。发现辣椒素以剂量和时间依赖性方式抑制FaDu细胞的生长和增殖。用50,100,200和300μM辣椒素处理的细胞随着辣椒素剂量的增加显示出细胞生长的增加的减少。此外,随着孵育时间的增加,活细胞的百分比降低。观察到的IC50值约为150μM[3]。

[体内研究]

辣椒素(CAP)处理的动物(组IV)显示增加的DNA片段化,表明与B(a)P-诱导的携带肺癌的动物(组II)相比显示细胞凋亡,其显示DNA片段化减少。与B组(a)P组给予第II组肺癌动物相比,CAP处理的IV组动物显示p53,Bax和caspase-3的表达显着增加,抗凋亡蛋白Bcl-2水平显着降低[ 4]。辣椒素在雌性Wistar/ST大鼠中引起剂量依赖性的泪液分泌减少。在20,50和100mg/kg的剂量下观察到显着效果。此外,辣椒素也会引起角膜病变,在50和100 mg/kg剂量下观察到显着效果[5]。

[细胞实验]

将FaDu细胞以1×10 5个细胞/孔的密度接种在24孔板上。过夜生长后,用不同浓度的辣椒素(0μM,50μM,100μM,150μM,200μM,250μM,300μM和350μM)处理细胞24,48和72小时,更换培养基每24小时一次。在处理结束时,添加30μL四唑鎓化合物MTT和270μL新鲜培养基。在37℃下进一步温育4小时后,向每个孔中加入200μL的0.1N HCl的10%SDS溶液以溶解四唑鎓晶体。最后,使用ELISA平板读数器记录540nm波长的吸光度[3]。

[动物实验]

小鼠[4]将体重20-25g(8-10周龄)的健康雄性瑞士白化小鼠分成四组,每组六只,每只小鼠如下。第I组:对照动物在整个实验过程中接受橄榄油。第II组:给动物施用苯并(a)芘(B(a)P)(50mg / kg体重,溶于橄榄油中),每周两次口服,连续四周。第III组:动物每周一次腹腔注射辣椒素(10mg / kg体重溶于橄榄油),持续14周,以评估辣椒素诱导的细胞毒性(如果有的话)。第IV组:动物腹膜内接受B(a)P(如第II组)以及辣椒素(10mg / kg b.wt溶于橄榄油中)。辣椒素治疗在第一剂B(a)P给药前一周开始,持续14周。大鼠[5]使用4日龄雌性Wistar / ST大鼠。在出生后第4天,给大鼠单次皮下注射辣椒素,剂量为50mg / kg(剂量反应试验为20和100mg / kg),溶于含10%乙醇和10%吐温80的生理盐水中。载体处理的大鼠单独接受载体溶液。注射的大鼠与其母亲一起饲养,母鸡喂食标准大鼠食物并保持在正常条件下。在4周龄时,将注射的大鼠分开并饲养在笼中,并保持在正常条件下直至实验开始。
